Why not just make some perk specific enemies? some that are hard to kill with certain weapon, but easy to kill with others.
your swarm idea is good, reminds me of the swarm of beetles in TSFP that came in a mass of a million crawling at the walls and floor of a long hallway. you would need a flamethrower or machinegun to kill them effectively, with singleshot weapons you would bound to die, forcing teamwork with other perks, this would complement the firebug and commando, be somewhat effective for the support, and pistol weilders, and be a nightmare for the sharpshooters.
now for another idea, maybe add an enemy that needs a headshot to kill, any other means would be innefective. and would regenerate health rather quickly. the contrary of a swarm, a single tough enemy with a small weak spot. complementing snipers/sharpshooters to kill it with they're strong one hit weapons.
some other zombie that takes no damage from fire, and becomes more dangerous if set on fire, a running torch might i say, maybe an increase in speed and damage while on fire
And another type wich would be almost impossible to kill from infront. give it some kind of boney armor and fast linear locomotion, with a weak spot from behind that does devestating damage when hit. (i took this idea from the prison zombie from Resident Evil 4.) forcing you to have a teammate wait on by when he rampages after you to shoot its weak back.
Maybe another one thats like a blob or spineless humanoid, that when hit by melee weapons it absorbs your weapon wich you can only get back when it dies. it could also phase throu doors and certain objects. grenades would be very effective since it would absorb it then blow up.
a certain specimen that runs after you and when it gets really close enough, opens up its body/torso to reveal teeth or some form of menacing tendrils, like a gaping mouth or so, berzerker and support would be effective since when it opens up its orifice to cause damage, you could just shoot it at close range or melee its weak tongue or other exposed frail organ.
i have no idea for the medic...
maybe some really diseased specimen that takes massive damage when hit by the syringe?...
