I would say Red Dead Redemption.
It was great fun to play with a good online mode. It had beautiful scenery which felt really organic and the storyline was crafted beautifully with believable and well crafted characters.

I haven't played Heavy Rain or Red Dead which both look excellent.
So my vote would go to Mass Effect 2 which is the most polished game of the rest. Mafia 2 and New Vegas come close but both have too many annoyances.
Another game which I enjoyed more than almost all on the list was Metro 2033

2010 was a bad year for PC gaming.
I can't see a single game that I would consider truly good, except Mass Effect 2 and SCII, which both don't cater to my tastes at all.

It was a pretty good year, only too bad Heavy Rain & RDR weren't released on PC, I'd love to play those. So I went with Mass Effect 2.

The original M&B was released in 2009. Warband in 2010.
SC2 hands down. Greatest RTS of all time. And best RTS campaign by far ever in terms of mission design (yes there were cheezy cutscenes).
And do I even need to mention the multiplayer? Utterly insane.
I loved CoH but I have to say I like SC2 even more. Plus CoH's SP sucked.
Second choice would be M&B:Warband.

Wasn't Call of Pripyat released 2010 (at least the English version)? But again, it was fun but not GOTY standard for me.
There's been a few good games this year, but no great games. Out of that list I was most interested in Mafia 2 but we know how that turned out. Want to play Mass Effect 2, and it would have probably been my pick, but some git stole my copy of Mass Effect 1 and I've yet to get/borrow another copy to finish it.
Still this year has given me more time to play through and pick up some older games* from the last 2-5years.
*That did say older gems, but I'm playing through Two Worlds right now and that is definitely not a gem.

I have not played FO NV yet, but that looks like another good game.
The rest are not even worth mentioning.
I can't stand the scale and "zoomed in" style of gameplay that SC seems to have.
Homeworld 2 is the best RTS of all time.
Someone needs to make a HW3 or similar game.
