Toonstruck (<- I loved that game.)

Not really a comedy game but I laughed loud quite a few times in those games:
Gothic 1 and #2 too sometimes. It's mainly the over-the-top voice acting in the original German version that makes it funny. Some universally funny bits too though.

There where quite a few back in the day, the Monkey Island series, Leisuresuit Larry, Grim Fandango, Day of the tentacle, and many more allready mentioned, like NOLF and Sam & Max.

But they are spread pretty thin thease days, i haven't played the new Sam & Max eppisodes i must admit, so i can't speak for thouse, but the last really funny game i player was probably Psychonauts, a game that easilly deserves to be on the list mind you!
